i have never had a problem going up a boat ramp and i have been up several. that is in 2wd. never put my my truck into 4wd on a boatramp.

Ramps are not standardized though they should be, so use your judgement.
At 50' I'm thinking you're not going by yourself so maybe a "helper" truck would be a back up.
Any ramp you NEED that for is a no-no IMO.
4WD would be a waste if towing that boat is the main function of that truck.
Just killing gas miliage, wasting power and more parts to break.

Consider leaving the boat in the water, renting it out as a condo when you're not using it
and take a 4WD someplace fun

That's a judgement call cuz only you know where you drive, just make sure you don't under-size the truck.
When you get the truck, you're taking us all fishing, right ?

not sure for anything that big. I would think it would have to be custom. Since it is a heavy boat and you are in search of a trailer it would be a good thing to look for a 5th wheel or gooseneck trailer or if it comes down to having one built that would be the way I go.
